Two relatively simple methods of good accuracy, precision, sensitivity and specificity are presented. Steroid conjugates from 50 ml nonpregnant urine are precipitated with ammonium sulfate (Cohen, 1966) and solvolyzed in ether perchloric acid. Free steroids are extracted with isopropyl ether (Touchstone, 1969). After phenolic separation and a single thin layer chromatography gaschromatographic quantitation of free or acetylated estrogens is performed. In the second method the diazonium derivatives are separated on a thin layer plate and quantitated as such by densitometry.
